All-rounder Ryan Stevenson is making his comeback from injury but did not bowl as Bradley Erasmus (136*) and Gary Wilson (96) dominated the day's batting.
Brad Wheal (2-86) was the pick of the Hampshire attack.
Surrey pace prospect Matt Dunn was the start of the day with the ball, finishing with figures of 5-1-8-5
Hampshire U17 batsman Billy Mead (no relation to the county's record run scorer Phil Mead) is making his 2nd XI debut and was 3 not out at the close.
